蝾螈迟钝爱德华菌的分离鉴定 被引量:3

摘要 为确定引起陕西某水产养殖场发病蝾螈的病原菌,从发病蝾螈的体表脓肿组织中采用细菌分离、生物学特性观察、生理生化试验、16SrRNA基因测序、人工感染试验和药敏试验对病原进行分离鉴定。结果从发病蝾螈体表脓肿组织中分离培养获得1株革兰氏阴性小杆菌,根据生物学特性、生理生化试验等初步鉴定为迟钝爱德华菌,通过16SrRNA序列(GenBank登录号JX570593)测定,在GenBank上经同源性序列比对,与迟钝爱德华菌(登录号JF699759)聚为一类,与大部分迟钝爱德华菌的16SrRNA的同源性均在98%以上。该菌对小白鼠有致病作用,可引起小白鼠皮肤肿胀、化脓,肌肉苍白、溃疡;蝾螈背部注射该菌后造成局部化脓、坏死或溃疡;该菌对庆大霉素、氧氟沙星、诺氟沙星等抗菌药物敏感,对头孢哌酮、红霉素、四环素等抗菌药物有耐药性。综合生理生化指标、分子生物学鉴定及动物致病性试验结果,确定分离菌株为蝾螈源迟钝爱德华菌。 A small Gram-negative bacterium,SX-Et11 ,was isolated from the samples of diseased sala- mander from an aqua farm in Shaanxi Province. The isolated strain was identified as Edwardsiella tarda based on the biological characteristics, biochemical tests and 16 S rRNA gene sequence analysis. The 16 S rRNA sequence homology of the isolated strain was above 98%compared with these of most E. tarda available in GenBank. The animal experiments showed that the isolated strain caused white mice skin swel ling and festering,and muscle pale and ulcer. It also caused local maturate,necrosis and ulcer in the injection site of salamander. The isolated strain was sensitive to gentamicin, ofloxacin, norfloxacin, and so on, while it was resistant to cefoperazone, erythromycin, acheomycin, etc. Based on the results obtained, the strain SX-Et11 was identified as salamander E. tarda.
