长期高嘌呤饮食诱导高血压动物模型 被引量:11

A novel hypertensive rat model induced by long-term high-purine diet

摘要 目的:饮食不节可能会影响人体生化指标异常,采用动物长期高嘌呤饲料喂养法,以期得到模拟人类不良生活方式诱导的高血压动物模型。方法:采用SD大鼠长期高嘌呤饲料(普通饲料、酵母干粉、腺嘌呤按一定配比制成)喂养,每天正常饮水,连续30周建立接近人类病症的高血压大鼠模型。于造模0、8、15、17、21、30周分别采用无创尾动脉法测定大鼠收缩压(SBP)、舒张压(DBP)、平均动脉压(MBP);于8、10、12、18、20、24周取血,测定血清丙氨酸转氨酶(ALT)、天门冬氨酸转氨酶(AST)、γ-谷氨酰转肽酶(GGT)、碱性磷酸酶(ALP)、总胆固醇(TC)、甘油三酯(TG)、尿酸(UA)、肌酐(Cr)水平;于7、9、11、16、19、22周进行尿代谢,测尿Cr、UA水平。结果:大鼠高嘌呤饲料喂养8周时,以收缩压为主,血压开始明显升高,17周时收缩压接近达到160mmHg;大鼠高嘌呤饲料喂养18周时血清ALT、AST、ALP、GGT、UA、Cr水平升高;11周时尿液UA水平降低;16周时尿液Cr水平降低,提示肝肾功能生化指标出现紊乱。实验中大鼠血清TC、TG未有显著差异,提示长期高嘌呤饲料喂养对大鼠血脂可能未有影响。结论:采用长期高嘌呤饲料喂养造模法,连续造模17周以上能使SD大鼠的血压升高到一定程度,达到轻、中度高血压水平,同时还能造成肝肾损伤等伴随症状。 Objective:Improper diet may lead to the biochemical criterion disorder of humans,SD rats were fed with high-purine diet to develop a novel life-habitual hypertension model,which could closely mimic the human diseases.Methods:To build the rat model of life-habitual hypertension,SD rats had free access to high-purine diet(chow,yeast powder,adenine mixed in a certain percentage) and tap water for 30 weeks.The non-invasive blood pressure(including SBP,DBP,and MBP) of conscious rats were measured directly by tail arteries in 0,8,15,17,21,30 weeks.ALT,AST,GGT,ALP,TC,TG,UA,Cr activity in serum were detected in 8,10,12,18,20,24 weeks.Cr,UA activity in urine were detected in 7,9,11,16,19,22 weeks.Results:After 8 weeks,the blood pressure of SD rats was started to significantly increase and the SBP was close to 160mmHg after 17 weeks.ALT,AST,ALP,GGT,UA,Cr activity in serum was increased(P0.01~0.05) after 18 weeks;UA activity in urine was reduced(P0.01) after 11 weeks and Cr activity in urine was reduced(P0.01) after 16 weeks,which indicated functional lesion of liver and kidney.There was no significant change about TC and TG levels in serum,indicated that the long-term high purine diet has no influence on the blood lipid level of SD rats.Conclusion:Long-term high-purine diet can increase the blood pressure of SD rats,which led to mild-to-moderate hypertension,and cause functional lesion of liver and kidney after more than 12 weeks.
