摘要
目的:研究维药复方薰衣草糖浆对D-半乳糖(D-gal)致老年痴呆模型小鼠的保护作用。方法:NIH小鼠120只,随机分为六组:正常对照组(NC),模型组(D-gal),维生素E片组,复方薰衣草糖浆低、中、高三个剂量组:分别为4.5ml/kg、9.0ml/kg和18.0ml/kg(分别相当于含生药量0.67g/kg、1.35g/kg和2.70g/kg)。每组小鼠20只。除NC组外,各组小鼠均颈背部iH D-gal50mg/kg。维生素E片组和复方薰衣草糖浆3个剂量组均ig给药,D-gal组和NC组小鼠分别ig等容量生理盐水,每日一次,造模和给药均连续60天。分别以跳台实验、迷宫实验法测定其潜伏期和5min内错误次数,并测定各组小鼠脑组织SOD活性及MDA含量。结果:跳台实验和迷宫实验中,D-gal组潜伏期和5min错误次数均明显高于NC组和各给药组,给药各组均可使潜伏期和5min错误次数明显降低。模型组小鼠脑SOD活性低于、MDA含量则高于NC组,用复方薰衣草糖浆均可使SOD活性明显提高,MDA含量明显降低。结论:复方薰衣草糖浆能显著提高老年痴呆模型小鼠的学习、记忆能力,提高脑组织SOD活性,降低脂质过氧化产物MDA的含量。
Objective:To investigate the protective influence of compound lavender syrups on D-galactose induced senile dementia model mice.Methods:120 NIH mice were randomly divided into six groups(20 per group):normal control group(NC),model group(D-gal),Vitamin E tablets groups,compound lavender syrups groups which were divided into low-,middle-and high-dose:4.5ml/kg、9.0ml/kg and 18.0ml/kg(equivalent to the content of crude drug 0.67g/kg、1.35g/kg and 2.70g/kg respectively).Except for NC group,other groups were given hypodermic injection of D-gal(50mg/kg).NC group was given hypodermic injection of NS.Vitamin E tablets group and compound lavender syrups groups were given oral administration.NC group and D-gal group were given oral administration of water.Mice of every group were repeatedly given hypodermic injection and oral administration drugs one time every day.Respectively measured erroneous number in 5 minutes,latent periods,SOD activities,and MDA content after 60 days.Results:Compared with other groups,D-gal group were obviously higher on erroneous in 5minutes which were 11.20±3.10 and 24.19 ±13.01(P0.05 or P0.01).Compared with other groups,D-gal group were obviously increased latent periods which were 35.61±8.64s and 53.75 ±17.85s(P0.05 or P0.01).Compared with other groups,D-gal group were obviously decreased SOD activities and increased MDA contents(P0.05 or P0.01).Conclusion:Our results indicated that compound lavender syrups could improve the function of learning and memory of the senile dementia model mouse,increase SOD activities and decrease MDA of brain.
