摘要
建立了大豆分离蛋白中三聚氰胺的液相色谱方法。大豆分离蛋白经过1%三氯乙酸和饱和乙酸铅溶液提取后,经过阳离子交换固相萃取柱净化处理,用5%氨化甲醇洗脱,洗脱液用高效液相色谱紫外检测器检测,外标法定量。三聚氢胺的质量浓度在0.4~40mg/L范围内与色谱峰面积呈良好的线性关系,相关系数为0.9990。方法的检出限为0.6mg/kg,定量限为2mg/kg。2,4,20mg/kg3个浓度水平的加标回收率在78.82%-92.95%范围内。该方法分析成本低,测定结果准确,能满足常规检测及食品安全监控的需要。
Liquid chromatography was developed for determining melamine residues in soy protein isolate. The target compound was extracted by trichloroacetic acid and saturation of lead acetate and cleaned up with cation-exchange solid phase extraction column. The extract was eluted with mixture of amonium methanol (volume ratio was 5 : 95). The determination was performed on LC with UV detector and the external calibration standard was used for quantification.The melamine concentration was linear with peak area in the range of 0.4-40 mg / L with r2=0.999 0. The detection limit was 0.6 mg/ kg, the quantification limit of melamine was 2 mg/kg. The recovery at levels of 2,4,20 mg/kg ranged from 78.82% to 92.95%. This method provids accurate results with less time consuming and lower cost, which can be meet the requirements of the routine testing and food safety monitoring.
