养肝和血方对卵巢早衰大鼠性激素的影响 被引量:9

摘要 目的通过观察养肝和血方对雷公藤多甙片所致的卵巢早衰大鼠相关指标的影响,探讨其治疗卵巢早衰的疗效和作用机制。方法取40只动情周期规律的雌性Wistar大鼠,随即分为空白组(n=10),造模组(n=30),用雷公藤多甙片灌胃法建立大鼠卵巢早衰动物模型,造模成功后,随机分为养肝和血方实验组(n=10)、己烯雌酚对照组(n=10)、模型对照组(n=10),分别给予养肝和血方溶液、己烯雌酚溶液、模型对照组和空白组给予等量生理盐水,连续灌胃15d。用阴道脱落细胞涂片法观察大鼠动情周期,放射免疫法检测大鼠促卵泡成熟素(FSH)、促黄体生成素(LH)、雌二醇含量(E2),HE染色法观察大鼠卵巢组织形态,统计方法采用SPSS19.0。结果养肝和血方可提高卵巢早衰大鼠血清E2的含量,降低大鼠FSH、LH含量,改善病理状态下卵巢组织的形态。结论养肝和血方从肝论治治疗卵巢早衰大鼠效果较好,和己烯雌酚具有相似的作用。 Objective By observing Related indicators changes of the premature ovarian failure caused by Yripterygium gavage .To explore the outcomes and mechanisms of Yangganhexue decoction in the treatment of premature ovarian failure. Methods Choose 40 female Wistar rats with regular sexual cycle .All rats were randomly divided into normal groups (n=10) and models group (n=30).Rat models of premature ovarian failure were established by Tripterygium garage They were subdivided into 3 groups, Yangganhexue decoction group, diethylstilbestrol control group and model control group, and then they were respectively treated with Yangganhexue decoction for 15 days and diethylstilbestrol for ~ 5 days, model control group and blank control group were managed with same amount of normal saline for 15 days. To confirm theanimal model ,vaginal smears were collected everyday for the rat estrous cycle.Using radioimmunoassay (RIA) of serum sex hormone estradiol (E2), follicle-stimulating hormone (FSH),luteinizing hormone (LH) levels. HE staining optical microscope observation of structural changes in the overies. Statistical Methods were used SPSS19.0. Results Yangganhexue reduced serum follicle-stimulating hormone (FSH) and luteinizing hormone (LH) levels, improved estradiol (E2) levels in rats with premature ovarian failure. Yangganhexue improve the Pathological state of ovarian morphology. Conclusion Yangganhexue has a similar role with diethylstilbestrol in the treatment &premature ovarian failure from the feasibility of the liver.
