摘要
目的 研究 Vit E和硒对人髓系白血病细胞株 HL-60细胞凋亡和 c-myc基因表达的影响。方法 应用体外细胞培养法 ,DNA凝胶电泳技术、Northern印迹杂交技术。结果 Vit E( 10 0 μmol/L)和 Se( 8μmol/L)作用 2 4h后 ,HL-60细胞出现细胞凋亡现象。Vit E( 10 0 μmol/L)可以抑制 c-myc基因的表达 ,作用有显著性 ( P <0 .0 5 )。Se( 8μmol/L)无此作用。结论 抑制 c-myc基因的表达 ,诱导白血病细胞凋亡是 Vit E抑制白血病细胞增殖的重要途径之一。硒与 Vit E同样具有抗氧化作用 ,可诱导 HL-60细胞凋亡。但对 c-myc基因的表达却无影响 。
Objective To study the effects of vitamin E and selenium on the induction of apoptosis and c myc gene expression in human leukemia cells.Methods DNA gel electrophoresis and Northern blotting hybridization were used to detect the apoptosis and the level of c myc gene,respectively.Human leukemia cell line HL 60 was cultured in vitro.Results The apoptosis of HL 60 cells was induced after exposure to vitamin E (100 μmol/L) and selenium (8 μmol/L) for 24 hours,respectively.C myc mRNA was down regulated significantly by vitamin E (100 μmol/L),but not observed in selenium (8 μmol/L).Conclusion Our observations suggested that c myc down regulation and induction of apoptosis by vitamin E is one of the important pathway to repress leukemia cell proliferation.Selenium can induce apoptosis of leukemia cells but not effect the expression of c myc gene.The results suggest there are different mechanisms in repressing leukemia cell proliferation between vitamin E and selenium.
