摘要
测定了猪源罗伊氏乳杆菌生长特性和抗逆特性,采用全自动曲线分析仪测定了该菌的生长曲线,筛选出其最适培养温度和pH值,探讨了在不同时间、不同温度下该菌的耐热特性,模拟了在动物胃肠道环境下,该菌对人工胃液和人工肠液的耐受性.结果表明:在37℃、pH 7.0的培养条件下,该菌生长情况最好,活菌数可达到109 CFU/mL以上;该菌培养3h进入对数生长期,培养19h进入生长稳定期.经过65℃的高温处理1min,该菌活菌数仍可达到109 CFU/mL以上.在人工胃液(pH 3.0)和人工肠液中培养2h,该菌的存活率在90%以上;处理4h时,存活率仍达到78.13%.说明该罗伊氏乳杆菌具有良好的生长特性和抗逆特性,可望成为饲用益生菌,服务于养猪生产.
To explore new feed probiotics strain,the growth characteristics and stress resistance of porcine Lactobacillus reuteri was evaluated in this study. The strain's growth curve was measured using automatic curve analyzer, the optimum culture temperature and pH were screened, and the heat resistance at different temperature was detected. And the tolerance to artificial gastric juice and artificial intestinal juice were estimated through the simulation of animal gastrointestinal environment. The results showed that the optimum growth temperature and pH were 37 ℃ and 7.0 with the viable count achieved 109 CFU/mL. This strain entered logarithmic phase and stationary phase within 3 h and 19 h, respectively. The survival rate was 76.82% when sustained at 65 ℃ for 1 min. Under the artificial gastric juice (pH 3.0) and artificial in testinal juice,the survival rate was above 90% for 2 h,while the rate was 78. 13% for 4 h. It was suggested that Lactobacillus reuteri strain had a good growth characteristics and stress resistance,and could be considered as a potential probiotic for improving pig production.
