摘要
利用噬菌体7肽库筛选出aFGF特异性结合噬菌体并鉴定其活性.经过四轮淘选,ELISA检测结合活性,然后挑取单克隆测序.竞争抑制试验验证阳性克隆与aFGF的结合活性.MTT法检测阳性噬菌体对aFGF诱导的Balb/c3T3细胞增殖的影响.一系列实验结果显示,筛选出的P8号阳性噬菌体与aFGF结合的特异性很高,对aFGF诱导的Balb/c3T3细胞增殖具有抑制作用.通过本实验,获得了具有与aFGF高度结合的噬菌体短肽,可为抗肿瘤新药提供重要基础.
The specific binding phage by using phage display peptide library and its activity was certified. After four rounds of screening, ELISA was applied to assess the combining activity, and the positive clones were picked out to sequence. The combining activity of positive clone and aFGF was examined by competing inhibition experiments. The influence of positive clones on the inhibition of Balb/c3T3 cell increment induced by aFGF was tested by MTT. A series of results figured that the specificity of the No.P8 positive phage which combine with aFGF was high. It could restrain the Balb/c3T3 cell proliferation induced by aFGF. Through this experiment, we have gain a phage peptide which could combined with aFGF highly.h could be a new drug for the tumor.
