摘要
目的 :建立检测心肌、肝、肾、肠组织中阿霉素含量的反相高效液相色谱法及初步应用于大鼠阿霉素肝血管输注 2 h后心肌、肝、肾肠组织中阿霉素含量的检测。方法 :取组织 1 .0 g制成匀浆 ,经氯仿 -甲醇 ( 4:1 )混合液提取 ,流动相 :甲醇 -0 .0 1 M醋酸钠溶液 -冰醋酸 ( 70 :3 0 :1 )混合液 ( p H:8.6) ,流速 :1 .0 ml/ min,柔红霉素为内标。采用 wistar大鼠肠系膜静脉输注阿霉素 ( 1 5mg/ kg) ,2 h后处死大鼠 ,取心肌、肝、肾、肠组织检测阿霉素含量。结果 :心肌、肝、肾、肠组织中阿霉素线性范围分别为 0 .4~ 3 .5ug/ g、 0 .5~ 5.0 ug/ g、 0 .5~ 7.5ug/ g、 0 .5~ 4 .5ug/g,最低检测限为 0 .1 ug/ g,平均方法回收率分别为 :89.4 0 %、 90 .80 %、 83 .59%、 87.3 9% ;相对偏差分别为 1 0 .60 %、 9.2 1 %、 1 6.4 1 %、 1 2 .61 %。大鼠肠系膜静脉输注阿霉素 2 h后心肌、肝、肾、肠组织阿霉素含量分别为 4 .0 6± 0 .56、 2 .70± 0 .3 9、 5.95± 0 .62、 3 .3 2± 0 .2 8。结论 :反相高效液相色谱法检测心肌、肝、肾、肠组织中阿霉素含量快速、准确。
Objective:To establish the method of RP-HP C for measuring the adriamycin(ADM)content of heart、liver、kidney、intestine tissue and primarily applied in measuring the ADM content of heart、liver、kidney、intestine tissue after ADM rat hepatic vascular infusion 2hours.Methods:1 0g tissue was homogenized and extracted with chloroform-methanol,mobile phase:methanol-0 01 M sodium acetate -acetate (70:30:1) pH 8 6,flow rate:1 0ml/min,daunorubicin is internal standard.ADM 15mg/Kg were infused through the Wistar rats mesenteric vein. After 2hours,the rats were sacrificed and the content of heart、liver、kidney、imtestine tissue、were measured.Results:The calibration curves were linear over the ADM content range of 0 4~3 5ug/g、0 5~5 0ug/g、0 5~7 5ug/g、0 5~4 5ug/g heart、liver、kidney、intestine tissre respectively,the lowest limit of detection was 0 1ug/g,the method recover rate were 89 40%、90 80%、83 59%、87 39%、respectively,RSD were 10 60%、9 21%、16 41%、12 61%separately,after ADM rat hepatic vascular infusion 2 hours,ADM content in heart、liver、kidney、intestine tissue were 4 06±0 56、2 70±0 39、5 95±0 62、3 32±0 28 respectively.Conclusion:The method of RP-HPLC for measuring the adriamycin (ADM)content of heart、liver、kidney、intestine tissue is fast、convenient and accurate.
