摘要 目的研究Notch1在喉鳞状细胞癌(简称喉鳞癌)组织及体外培养Hep-2细胞中的表达及其与喉鳞癌病例临床病理资料之间的关系。方法应用量子点超敏免疫荧光(QDs immunofluorescence)染色法检测Notch1蛋白在95例喉鳞癌、31例声带息肉组织标本及Hep-2细胞中的表达。结果 Notch1在95例喉鳞癌组织中表达阳性率为88.4%,其表达主要定位于细胞质和细胞核。在喉鳞癌组织中Notch1的表达与T分级、临床分期和淋巴结转移有显著相关性。Notch1在喉鳞癌组织细胞核中的阳性表达与T分级、临床分期、病理分级和淋巴结转移有显著相关性。喉鳞癌细胞Hep-2中Notch1表达阳性,且主要定位于细胞质和胞膜中。结论 Notch1与喉鳞癌的发生发展有着重要关系,其作用的具体机制有待深入研究。 OBJECTIVE To investigate the expression of Notch1 protein in laryngeal carcinoma cell line (Hep-2) and the laryngeal carcinoma tissues, and to study the relationship between the expression and the clinical and pathological data in laryngeal carcinoma patients. METHODS The expressions of Notch1 in 95 laryngeal carcinoma tissue, laryngeal carcinoma cell line (Hep-2) and 31 vocal cord polyp specimen (as control group) were detected by QDs-lgG conjugate immunofluorescence staining. RESULTS The positive expression of Notch1 in cells of laryngeal carcinoma tissues was mainly localized in the cytoplasm and the nucleus. The expression rate of Notch1 in laryngeal carcinoma tissues was 88.4%, and it was significant higher than that in vocal cord polyp tissues. The expression of Notch1 in laryngeal carcinoma tissues was associated with its clinical stage, T classification and lymph node metastasis, and the positive expression of Notch1 in the nucleus of cells of laryngeal carcinoma tissues was related to T classification, clinical stage, pathological grade and lymph node metastasis. The expressions of Notch1 in laryngeal carcinoma cell line (Hep-2) was positive and mainly localized in the cytoplasm and membrane. CONCLUTION Notch1 may play an important role in the development of laryngeal carcinoma, and the mechanism need further study.
