摘要
作者使用兔肌肉组织,4℃下经匀浆、Ca_3(PO_4)_2凝胶吸附、0.2 mol/L pH 7.2磷酸盐缓冲液洗脱、硫酸铵及丙酮分级沉淀,再用CS-1实验型超滤过器,截留分子量(Mr)200000超过滤膜经氮气加压过滤,然后装入透析袋置4℃透析,8—10h等处理程序,提取纯化乳酸脱氢酶(LDH).纯化后的LDH活力为1602000IU/L,含蛋白7.59g/L,经初步应用,与进口LDH的结果基本一致,t=0.738,P>0.05,能满足试验的要求,可代替进口酶.
Lactate dehydrogenase(LDH )was extracted and purified from rabbit musde tissue at 4℃ by the process of homogenate , Ca3( PO4)gel adsorption , dution in 0.2 mol/L, pH 7.2 phosphate buffer , (NH4)2SO4, precipitation in classification with acetone , pressuring percolation passing through 200 000 (Mr ) ultrafilted membrance by N2 using CS -I lab ultrafiltor and dialysis at 4℃ for 8-10 hours , etc . The method is in low price . The activity of purified LDH was 1 602 000 IU/L , containing 7.59 g/L of protein . After application , it is proved to be equal to imported one from sigma (t=0.738, P>0.05 )and can fulfill the experiment requirement and replace the latter .
