摘要
作者用兔抗葡萄球菌肠毒素B(SEB)多克隆抗体作为包被抗体,以HRP标记的抗SEB McAb作为第2抗体,建立了一种McAb-ELISA夹心法,用于检测人工污染食物体本敏感性的下限为1.5μg/L.并比较了吐温20,BSA,低脂牛奶及8种正常血清等作为抑制剂消除试验中非特异性反应的效果.结果表明,羊或兔血清和吐温20的使用浓度分别以2% 10%和0.05%-0.15%为佳,且发现血清与吐温20还具有协同抑制作用.
A McAb-FLISA sandwich method was developed using polyclonal antibody (anti - SF.B in rabbit) as coating antibody , and horseradish peroxidasc ( HRP ) labeled monoclonal antibody as second (sandwich ) antibody for detecting food specimens artificially contaminated with SF.B . The lower limit of sensitivity of the method has reached 1.5 μg/ L . The effects of Tween - 20, bovin scrum albumin ( BSA ). low - fat milk and 8 species of normal sera used as inhibitors to remove non - specific reaction in the tests were compared . The results show that optimal concentrations of sheep or rabbit scrum and Twecn -20 are 2%-10% and 0.05%-0.15% respectively, and sug-gest that there arc synergistic inhibitory effects between the two sera and Twecn-20 mentioned above .
