摘要
作者用化学发光法、~3H-脱氧胸腺嘧啶核苷(~3H-TdR)参入DNA合成法和形态学方法,研究了蚯蚓提取物(912)对激光-血卟啉衍生物(HpD)的增敏作用及其机理。结果:发光强度(以每10秒脉冲计数表示):A组(S_(100)为364.2±42.8,做为对照;B组(laser-S_(100))为372.7±79.9,C组(laser-912-S_(100))为965.2±214.9,D组(912-S_(100))为1.188.8±387.7,E组(laser-HpD-S_(100))为1703.5±700.3,F组(laser-HpD-912-S_(100))为4072.8±2133.4,以F组发光强度为最高,与其它各组比较,差异有非常显著的意义(P<0.01)。细胞损伤亦以F组最重。结果表明:912与laser-HpD对癌细胞具有协同杀伤作用。如将自由基清除剂—过氧化氢酶(CAT)加入上述各管中,则发光强度降为对照管的70%-50%,CAT和SOD合用,各管降为10%-5%结果提示,被测体系中有过氧化氢(H_2O_2)和超氧阴离子自由基(O_2^-)产生。说明912对laser-HpD杀伤癌细胞的增敏作用至少部分是通过增加活性氧而实现的。
Aidiron (earthworm extract, 912) for sensitizing tumour cells S180 to HpD-laser light was studied through measurement cnemiluminescence, biochemical synthesis of DNA by incorporating 3 R-TdR and through observation of ultrastructure of tumour cells. The sensitization mechanism was also studied The results showed that the combined laser-HpD-912 treatment had the highest luminescent intensity (LI) (4072 pulse counts/10s) (P<0.01). The DNA formation was the lowest (7259 cpm) in tumours exposed to the combined laser-HpD-912 treatment, and the mortality of the tumour cells was the highest (74.1%), (P<0.01). Changes in the ultrastructure of the tumour cells were also the greatest This suggests that the toxic effects of HpD-laser on the tumour cells were enhanced by the use of 912 The LI of the experimental groups was 70%-50% of the control when CAT was added alone and decreased to 10%-5% when CAT and SOD was added simultaneously. Therefore, the toxic effect of laser-HpD on tumour cells by increasing concentration of free radicals and reactive oxygen.
