白藜芦醇对哮喘小鼠气道炎症干预作用的研究

摘要 目的探讨白藜芦醇对哮喘小鼠模型中气道炎症及相关细胞因子水平的影响。方法雾化吸入卵蛋白(OVA)-氢氧化铝[Al(OH)3]建立小鼠哮喘模型,雌性SPF级BALB/c小鼠18只,分为白藜芦醇组、阳性对照组和阴性对照组各6只。阿尔新蓝/过碘酸雪夫染色,观察黏液分泌情况,免疫组化检测基质金属蛋白酶9(MMP-9)、核因子кB(NF-кB)水平,ELISA检测血清白介素4(IL-4),γ干扰素(IFN-γ)和免疫球蛋白E(IgE)含量,瑞氏染色计数支气管肺泡灌洗液嗜酸性粒细胞。结果阳性对照组嗜酸粒细胞升高、黏液分泌增加,MMP-9及NF-кB蛋白表达,IL-4水平及IgE含量均明显增高(P<0.05);IFN-γ水平下降(P<0.05)。白藜芦醇组与阳性对照组比较,嗜酸粒细胞计数稍升高,MMP-9、NF-кB蛋白表达均降低,IL-4水平和IgE含量均降低;IFN-γ水平升高(P<0.05)。与对照组差异无统计学意义(P>0.05)。结论白藜芦醇干预可降低气道炎症,改变哮喘相关细胞因子水平。 Objective To evaluate the modulating effects of Resveratrol on the pathological features of airway inflammation and the expression of inflammation related multiple cytokines in asthmatic mice models. Methods BALB/e mice sensitized with ovalbumin conjuncted with AI(OH) 3 ,were challenged with repeated exposure to aerosolized ovalbumin. 18 SPF mice were divided to three teams : Resveratraol, positive and negative team. intedeukin-4 ( IL-4 ), immunoglobulin-E ( IgE ) γ Interferon-γ (IFN-γ) was measured by ELISA and the Aleian Blue/PAS staining was used to identify airway goblet cells. The expression of matrix metalloprotein- ase 9 (MMP-9) and neclear factor-KB (NF-KB) in bronchi and lung tissues were observed by immunohistoehemistry. Results After repetitive allergen challenge obvious infiltration of inflammatory cells and proliferation of goblet cells in mouse bronchus were observed. Expression levels of MMP-9 ,NF-I〈B in the epithelial cells of bronchi and IL-4 ,IFN-γ in the serum were significantly higher in asthmatic animals than those of control group. IgE was decreased in asthmatic animals. Compared with asthmatic group, there was mild inflamma- tion deposition and decreased expression of MMP-9,NF-kB ,IL-4,IgE in Resveratrol treated group. Conclusion Resveratrol intervention could inhibit airway inflammation by modulation of allergy related muhiple cytokines.
