人参皂苷Rg3抑制肺癌NCI-H1650细胞增殖作用研究 被引量:13

摘要 [目的]研究人参皂苷Rg3对肺腺癌NCI-H1650细胞增殖的抑制作用及对凋亡的影响。[方法]MTT法检测人参皂苷Rg3对NCI-H1650细胞增殖的抑制作用,Annexin-V-FITC/PI法检测细胞凋亡,Western blot法检测caspase-3、Bcl-2、Bax、survivin蛋白表达的情况。[结果]不同浓度人参皂苷Rg3能有效抑制NCI-H1650细胞增殖,且呈时间剂量依赖性(P<0.01)。20μmol/L人参皂苷作用NCI-H1650细胞24、48h,细胞凋亡率明显增加,与对照组相比,差异有统计学意义(P<0.01)。20μmol/L人参皂苷作用NCI-H1650细胞48h,Bcl-2、survivin蛋白表达降低、caspase-3、Bax蛋白表达增加,与对照组相比,差异具有统计学意义(P<0.01)。[结论]人参皂苷Rg3对肺腺癌NCI-H1650细胞增殖具有明显的抑制作用,呈时间剂量依赖关系,其机制可能与降低细胞Bcl-2、survivin蛋白表达,增加caspase-3、Bax蛋白表达,促进细胞凋亡有关。 PurposeTo investigate ginsenoside Rg3 inhibiting proliferation of lung cancer NCI-H1650 cells and its influence on cell apoptosis. Methods Inhibitive effect of ginsenoside Rg3 on proliferation of lung cancer NCI-H1650 cells was detected by MTT assay,and cell apoptosis was detected by Annexin-V-FITC/PI assay,expressions of caspase-3,Bcl-2,Bax,survivin proteins were detected by Western blot. Results Ginsenoside Rg3 with various concentrations effectively inhibited the proliferation of NCI-H1650 cells with a time-dose dependent manner (P0.01). Compared with the control group,ginsenoside Rg3 in a concentration of 20μmol/L significantly increased the apoptosis rate of NCI-H1650 cells in 24h and 48h (P0.01). Compared with the control group,expressions of Bcl-2 and survivin decreased after NCI-H1650 cells treated with ginsenoside Rg3 in a concentration of 20μmol/L for 48h,with significant difference between the two groups (P0.01).Conclusion Ginsenoside Rg3 could significantly inhibite the proliferation of NCI-H1650 cells with time-dose dependent manner,and the mechanism may be related to reduce Bcl-2,survivin proteins’ expressions,increase caspase-3,Bax proteins’ expressions,promote cell apoptosis.
