摘要
目的探讨阿霉素(adriamycin,ADR)化疗与热化疗(热疗联合化疗)对人肺腺癌细胞株A549细胞的抑制作用与细胞内阿霉素浓度的关系。方法不同浓度的阿霉素作用于体外培养的A549细胞,42.5℃作用30、60min后,37℃继续培养,然后用MTT法检测细胞的毒性作用,用流式细胞仪检测细胞内阿霉素浓度。结果化疗、热化疗对A549细胞均有抑制作用,与对照组比较差异有非常显著性(P<0.01),且均存在浓度、时间的依赖性;热化疗对细胞的抑制作用明显强于单纯化疗(P<0.01);热化疗组细胞内阿霉素的浓度明显高于单纯化疗组(P<0.01);各组细胞内药物浓度与细胞的抑制率呈直线相关(P<0.05)。结论阿霉素热化疗可以显著增强对A549细胞的抑制,其抑制作用与细胞内浓度有关。
Objective To explore the relationship between the intracellular drug concentration and the inhibition on human lung adenocarcinoma A549 cell line by chemotherapy and thennochemotherapy of ADR. Methods ADR of different concentrations was directly applied to A549 cells cultivated in vitro. Cells were maintained at 42.5℃ for 30 and 60 minutes, then grown at 37℃. The inhibition of cell growth was observed with MTF assay and and the intracellular concentrtion of ADR was detected by flow cytometry. Results Themotherapy and thermochemotherapy significantly suppressed A549 cells compared with control( P 〈 0.01 ). Both chemotherapy and thermochemotherapy had a time and eoncentration dependent suppressive effect on cells. Growth arrest of Thermochemotherapy - treated cells was more evident than those treated by themotherapy( P 〈 0.01 ). cytoplasmic concentration of were positive correlation between the inhiibition rate and the intracellular concentration of A549 cells in chemotherapy groups and thermochemotherapy groups ( P 〈 0.05). Conclusion Thennochemotherapy of ADR significantly increased the suppressive effect , which is related to the intracellular concentration of A549 cells.
