ILK在异丙肾上腺素诱导大鼠心肌肥厚组织中的表达 被引量:1

摘要 目的探讨心肌肥厚发生发展过程中ILK的表达特征。方法在大鼠背部皮下分别注射7、28和56 d异丙肾上腺素(ISO),判定心肌肥厚指标,利用免疫组织化学和免疫印迹技术,观察ILK在肥厚左心室肌中的表达。结果 ILK主要表达于心肌细胞胞膜和内皮细胞。与对照组相比,ISO组ILK阳性表达明显增强。随着造模时间的延长,肥厚心肌中ILK的表达含量逐渐升高。Western blot图像分析结果显示:ISO各实验组大鼠左心室心肌组织中的ILK蛋白表达带均强于对照组(P<0.01),7、28和56 d组间比较差异有统计学意义(P<0.05)。结论 ILK与心肌肥厚的发生和发展有关,心肌肥大越重,ILK含量增加越高。 Objective To explore ILK expression in cardiac cells during development of myocardial hypertrophy.Methods The isoprenaline(ISO) were injected to rat dorsal subcutis for 7 d,28 d and 56 d respectively,and then myocardial hypertrophic indexes were assessed.The techniques of immunohistochemisty and western blot were used for investigating ILK expression in left ventricular tissue.Results ILK was mainly located at myocardial cell membrane and endothelial cell.ILK was expressed obviously higher than that of control group,ILK expression and content in hypertrophic myocardial tissue were increased gradually along with extending of hypertrophic model.The image analysis result showed that strips of ILK expression in left ventricular tissue of model rat was stronger than that of control group(P〈0.01),There were significant deviation between 7 d,28 d and 56 d(P〈0.05).Conclusion There are correlativities between ILK and development of myocardial hypertrophy,the more myocardial hypertrophy is severe,the higher ILK content is much.
