摘要
目的:观察风湿宁胶囊(FSN)对胶原诱导性关节炎(CIA)大鼠滑膜细胞的促凋亡作用并探讨其作用机制。方法:制备CIA大鼠模型,将大鼠随机分为正常组、CIA模型组、雷公藤多苷组、FSN低、中、高剂量组。采用ELISA法检测滑膜细胞色素C(Cytochrome C)含量,免疫组化法检测滑膜中B细胞淋巴瘤-2(Bcl-2)的表达,Western Blot法分析FSN对滑膜含半胱氨酸的天冬氨酸蛋白水解酶-3(Caspase-3)蛋白表达的影响。结果:FSN能够明显升高Cytochrome C水平(P<0.05),下调Bcl-2蛋白的阳性表达,促进Caspase-3蛋白的表达(P<0.05)。结论:风湿宁胶囊能诱导CIA大鼠滑膜细胞凋亡,其促凋亡机制可能与抑制Bcl-2表达,促进Cytochrome C释放入胞浆,激活Caspase-3介导的细胞凋亡线粒体途径有关。
Objective: To investigate the pro-apoptosis effect and mechanism of Fengshining Capsule (FSN) on collagen-induced arthritis (CIA) in rats synoviocyte. Methods: Rats were divided into normal comparison group, CIA model group, tripterygium glycosides group, FSN low dose group, FSN middle dose group and FSN high dose group. The content of Cytochrome C in synovium of rats was detected by ELISA assay, and its expression of Bcl-2 in syuoviocyte was detected by immunohistochemical assay. Caspase-3 protein expression was measured by Western Blot. Results: FSN significantly increased the levels of Cytochrome C (P〈0.05), and reduced the expression of Bcl-2 protein. Meanwhile, the expression of Caspase-3 was promoted too (P〈0.05). Conclusion: Fengshining Capsule might induce synoviocyte apoptosis in CIA rats. Its mechanism may be related to inhibiting the expression of Bcl-2, releasing Cytochrome C into the cytoplasm and activating mitochondrial pathway of apoptosis-related by Caspase-3 mediated.
