摘要
目的:探讨运脾和络颗粒对糖尿病周围神经病变(DPN)大鼠模型坐骨神经组织髓鞘碱性蛋白的影响。方法:造模大鼠随机分成正常组、模型组、运脾和络颗粒高、中、低剂量组、糖脉康组、甲钴胺组,采用免疫组化与实时荧光定量PCR法检测运脾和络颗粒对糖尿病周围神经大鼠坐骨神经组织髓鞘碱性蛋白表达的影响。结果:运脾和络颗粒高、中剂量组能明显上调模型大鼠坐骨神经组织髓鞘碱性蛋白的基因与蛋白表达(P<0.05)。结论:运脾和络颗粒能够上调DPN大鼠髓鞘碱性蛋白的表达,从而改善DPN的脱髓鞘病变。
Objective: To explore the effect of Yunpi Heluo Granule on Myelin basic protein(MBP) of the sciatic nerve in rats with peripheral neuropathy diabetic. Methods: The rats were randomly divided into the control group, the model control group, the high, medium and low dosage of Yunpi Heluo Granule groups, Tangmaikang Granule group, western medicine (Methylcobalamin) control group. The method of immunohistochemistry and PCR were used to observe the effect of Yunpi Heluo Granule on MBP in rats with peripheral neuropathy diabetic. Results: Compared with the model group, the protein expression of tissue MBP and gene expression of interving groups of Yunpi Heluo Granule increased to varying degrees, which medium-dose group was significantly higher and followed by high-dose group(P〈0.05). Conclusion: Yunpi Heluo Granule could up-regulate the expression of MBP in peripheral neuropathy rats, thereby improve the demyelinating lesions of diabetic peripheral neuropathy.
