摘要
首次应用高速逆流色谱法分离纯化茶黄素单体成分 ,溶剂系统为乙酸乙酯 -正己烷 -甲醇 -水 ( 3∶ 1∶ 1∶ 6) ,优化了分离茶黄素的条件。同时与 Sephadex L H- 2 0柱色谱法梯度洗脱对比 ,结果表明 ,高速逆流色谱法分离时间相对较短 ,可进行较大量的分离制备。高速逆流色谱法较之 Sephadex LH- 2 0柱色谱法还有一个突出的优点 ,即无不可逆吸附污染及不会导致样品化学变性。
The separation and purification of theaflavins on high speed countercurrent chromatography was observated.The result showed that the stationary phase was able to be effectively retained and theaflavins could be effectively separated into three parts under the conditions of 800 rpm,≤2.0 ml/min,≤250 mg injection amount in the solvent system[ethyl acetate/hexane/methanol/water(3∶1∶1∶6)].Comparative studies showed that Sephadex LH 20 gel chromatography was suitable for separation of small amount sample,and required more time,high speed countercurrent chromatogrphy for separation of relatively large amount sample and required less time.High speed countercurrent chromatogrphy had a special merit of no adsorption without a porous matrix to retain a stationary liquid phase.
