摘要
采用聚丙烯酰胺凝胶垂直板电泳法 ,对中国孤雌生殖卤虫 1 0个品系的 2 6个多倍体单个体克隆的 9种同工酶的基因表达情况、种群遗传结构以及进化关系进行了研究。在研究的 9种同工酶中LDH、 6PGDH、GDH为单态酶 ,MDH、POD、EST、ACP、ALP和α AMY为多态酶。MDH、POD重复基因不表达 ,为功能二倍体 ,EST、ACP、ALP和α AMY重复基因表达。以遗传相似系数所做的聚类分析结果可以明显表明中国孤雌生殖卤虫各品系多倍体克隆可被分为二个类群 :一个为山东、河北品系类群 ;另一个为辽宁、新疆、青海类群。各克隆的遗传相似系数 (I)值较高 ,说明亲缘关系较近。各品系多倍体克隆间存在多态现象 ,杂合度值较高 ,说明各品系多倍体克隆间遗传变异的程度较高。
The gene expression of nine isozymes (lactate dehydrogenase, glutamic dehydrogenase, 6 phoshogluconate dehydrogenase, peroxidase, esterase, acid phoshatase, alkaline phosphatase, malate dehydrogenase, α amylase)and population genetic structure of 26 clones of Artemia parthenogenetica polyploids from China were studied with polyacrylamide gel electrophoresis. The experimental results showed that:(1)LDH, 6PGDH and GDH were monomorphic, while MDH, POD, EST, ACP, ALP and α AMY are polymorphic;(2)Duplicate genes of MDH and POD were not expressed, and they were functional diploidization, and duplicate genes of EST, ACP, ALP and α AMY were expressed;(3)Cluster analysis by genetic identity showed that 26 clones in ten parthenogenetic Artemia strains from China could be divided into two subgroups: group of Shandong and Hebei and group of Liaoning, Xinjiang and Qinghai;(4)Polymorphism and genetic identity are relatively higher in 26 polyploids clones of Artemia strains than other animals.
