摘要
目的探讨miR-143在结直肠癌中的表达情况及其与临床病理指标间关系。方法采用RT-PCR法检测miR-143在17例结肠癌患者的肿瘤标本(结肠癌组)及癌旁黏膜组织(结肠黏膜组)和81例直肠癌患者的肿瘤标本(直肠癌组)及癌旁黏膜组织(直肠黏膜组)标本中的表达情况,并分析其与结直肠癌患者临床病理指标间关系。结果结肠癌组miR-143表达水平(Ct值为15.96±1.76)明显低于结肠黏膜组(Ct值为14.34±3.44),差异有统计学意义(P=0.023),直肠癌组miR-143表达水平(Ct值为14.66±3.18)与直肠黏膜组(Ct值为14.29±4.82)比较,差异无统计学意义(P=0.351);直肠癌组miR-145表达下调在性别、年龄、TNM分期、脉管浸润上差异无统计学意义(P>0.05)。结论 miR-143仅在结肠癌中表达降低,在直肠癌中表达无变化,miR-143仅参与了结肠癌的发生,结肠癌和直肠癌可能有不同的癌变分子基础。
Objective To investigate the expression of miR-143 and analyze its relationship with clinicopathological variables in colorectal cancer patients.Methods Real-time PCR was used to detect the expression of miR-143 in 17 cases of colon cancer and 81 cases of rectal cancer and the corresponding normal mucosa tissue.The relationship of the expression of miR-143 with clinicopathological variables including sex,age,TNM stage and vessel invasion was analyzed.Results The expression of miR-143 was lower in colon cancer tissue(Ct:15.96±1.76) than that in colon mucosa tissue(Ct:14.34±3.44)(P=0.023),and there was no significant difference between rectal cancer tissue(Ct:14.66±3.18) and rectal mucosa tissue(Ct:14.29±4.82)(P=0.351).There was no significant difference in miR-143 expression in different sexes,ages,TNM stages and vessel invasion(P0.05).Conclusion The expression of miR-143 is low in colon cancer tissue and does not differ in rectal cancer tissue,and is involved in the development of colon cancer.Colon cancer may have the different etiology from rectal cancer.
