摘要
目的 观察限食对仔鼠胸腺细胞的影响及其与细胞凋亡的关系。方法 本研究采用 Wistar大鼠从怀孕 1 6天进行限食喂养直到生后 45天 ,进食量分别为对照组的 50 % ,观察限食对仔鼠胸腺细胞凋亡及凋亡有关基因 Bax、Fas的影响。结果 限食组仔鼠死亡率增加 ,并出现胸腺皮质萎缩、髓质囊性变有细胞碎片等病理改变 ,胸腺细胞凋亡数 ( 2 7.0 % )明显高于对照组 ( 7.8% ,P<0 .0 1 )。凋亡细胞主要位于胸腺皮质及皮髓质交界处 ,且此处 Bax及 Fas蛋白的表达也明显增高 ,结论 限食可使仔鼠胸腺萎缩 ,凋亡增加 ,机体细胞免疫功能低下 。
Objective: It was to observe the effect of diet restriction on thymocytes and its relationship to apoptosis in offsprings. Methods: Pregnant rats were restricted half in dietary intake from day 16 postpregnancy to day 45 postpartum. Its effects on apoptosis and related genes (Bax and Fas) expression in the thymocytes were observed using TUNEL and immunohistochemistry methods. Results: The weight in diet restricted group was significantly decreased and appeared cortex atrophy. The percentage of apoptosis in the thymus was higher in diet restricted group (27%) than in control group (15.3%, P<0.01). Diet restriction could significantly increase the expression of Fas and Bax genes. Conclusion: Diet restriction in the early life may lead to thymus atrophy, increased apoptosis and result in decreased cellular immune function in offsprings. [WT5”FZ]
