麦胚黄酮类提取物诱导乳腺癌细胞株凋亡的作用 被引量:13

摘要 目的 检测麦胚黄酮类提取物对人体乳腺髓样癌细胞株 BCap- 37凋亡的诱导作用 ,探讨其分子学机制。方法 用流式细胞仪检测麦胚黄酮类提取物对人体乳腺髓样癌细胞株BCap- 37细胞凋亡的诱导作用及对 bcl- 2基因蛋白表达的抑制作用。用光学显微镜观察凋亡细胞的形态学特征。结果  1 .麦胚黄酮类提取物主要阻断乳腺癌细胞由 G2 - M向 S期转变 ,并能诱导细胞发生凋亡 ,呈现剂量 -效应关系 ( P<0 .0 0 0 1 )。 2 .麦胚黄酮类提取物亦能明显抑制人体乳腺癌细胞株 BCap- 37中 bcl- 2基因的蛋白表达 ,其表达率与麦胚黄酮类提取物浓度成负相关 ( P<0 .0 1 )。3.显微镜下观察 ,可见乳腺癌细胞出现凋亡特征性形态学改变 :细胞体积缩小、胞浆浓缩、胞核固缩、染色质靠核膜凝集成不同形态 ,如马蹄型、月牙型、肾型 ,形成凋亡小体。结论 麦胚黄酮类提取物具有诱导人体乳腺髓样癌细胞株 BCap- 37凋亡的作用 ,bcl- Objective: This study was designed to determine whether the flavonoids extract of wheat germ could induce breast cancer cell apoptosis and illustrate the possible molecular mechanism. Methods: Apoptosis of the cell line BCap 37 was detected by morphological observation and flow cytometric cell cycle analysis. The bcl 2 expression was determined by using flow cytometry. Results: 1.The flavonoids extract of wheat germ could induce a dose dependent cell apoptosis (P<0.0001) and selectively block cell cycle progression from G 2 M phase to S phase. 2.The flavonoids extract of wheat germ could inhibit bcl 2 expression in a breast cancer line BCap 37, and the expression was negatively correlated with the concentration of flavonoids extract of wheat germ (P<0.01). 3.The characteristic morphology of apoptosis cells included marked condensation of cytoplasm and nucleus, accompanied by a decrease in cell volume, and the nuclear chromatin was aggregated into unique shapes such as horse shoe, semi moon, kidney beneath the nuclear envelope, and finally formed apoptosis bodies. Conclusion: Flavonoids extract of wheat germ can induce apoptosis of the cell line BCap 37 partially through inhibiting the bcl 2 expression. [WT5”FZ]
