摘要
目的 探讨年龄对二甲苯神经毒作用的影响。方法 采用 H E 染色法和 T U N E L 技术对不同月龄大鼠的脑组织病理形态学和神经细胞凋亡情况进行了研究。结果 二甲苯500 m g/kg 染毒后,2 月龄组较8 月龄组大鼠脑组织形态学改变相对严重,2 月龄组染毒大鼠神经细胞凋亡的发生率为(1 .53 ±1 .06) 个/ 视野,明显高于同龄对照组的(0 .93 ±0 .75) 个/ 视野, F= 12 .659 6 , P< 0 .001 ;8 月龄组染毒大鼠的神经细胞凋亡率为(1 .65 ±1 .11) 个/ 视野,明显高于同龄对照组的(1 .15 ±1 .07) 个/ 视野, F= 6 .263 3 , P< 0 .05 。
Objective To evaluate the neurotoxic effects of xylene on m orphology and apoptosis ofneuronsin rats’brain ,and whether there could be any relation between the two groups of rats with differentages and the neruotoxic effects of xylene . Methods Male S Drats ,with age of 2 m onth or 8 month ,were ad ministered xylene intraperitoneally on alternate days at a dose of 0 ,125 ,250 or 500 m g/kg for 30 days . H Estaining m ethod and T U N E Ltechnique were used to examine the morphology of brain and the apoptosis ofneruons. Results The degeneration of neurons was observed both in brains of 2 month and 8 month in thethree exposure groups . Gliosis was detected in brain of 2 m onth at 250 and 500 m g/kg and in 8 m onth at 500mg/kg only . Oede ma and cellular infiltration were only seen in 2 month rats . The characteristics of T U N E Lpositive neurons are nuclear shrinkage and stronger fluorescence in nuclei than that in normal neurons . Thenum ber of T U N E Lpositive cells was 1 .53 per field of vision in exposed group and 0 .93 per field of vision incontrol group in brains of 2 month rats( P < 0 .001) . More T U N E Lpositive cells were seen in exposed group(1 .65 per field of vision) than those in control group(1 .15 per field of vision) of 8 month rats( P < 0 .05) . Conclusion Xylene induced m ore serious m orphological changes and more apoptosis cells in the brain ofyounger rat than in the brain of older one . Itis suggested that younger ratis more susceptible to the neurotoxiceffects of xylene than older one .
