摘要
目的:应用蛋白质组学技术寻找妊娠期高血压疾病诊断相关的尿液差异蛋白,为早期诊断提供依据。方法:收集妊娠期高血压疾病(妊娠期高血压、轻度子痫前期、重度子痫前期孕妇尿液样品各10例)及正常孕妇的尿液样品10例,进行双向凝胶电泳,利用ImageMaster2D软件分析结果,寻找差异蛋白并进行基质辅助激光解吸电离飞行时间质谱(MALDI-TOF-MS)鉴定。结果:对比分析妊娠期高血压疾病孕妇和正常孕妇尿液,共获得65个差异蛋白点,质谱鉴定出30种差异蛋白。其中妊娠期高血压与正常孕妇比较鉴定出22种差异蛋白,表达上调的有14种,下调的有8种;轻度子痫前期与正常孕妇比较鉴定出26种差异蛋白,表达上调的有5种,下调的有21种;重度子痫前期与正常孕妇比较鉴定出25种差异蛋白,表达上调的有3种,下调的有22种。结论:应用蛋白质组学技术成功筛选出妊娠期高血压疾病与正常孕妇尿液的差异蛋白,这些蛋白可能与妊娠期高血压疾病的发生、发展有关,为进一步寻找尿液中早期诊断该病的特异分子标记物提供了基础资料。
Objective:To screen the differential proteins between patients with hypertensive disorder complicating pregnancy and healthy pregnant women by two-dimensional gel electrophoresis and to provide a ba- sis for early diagnosis of hypertensive disorder complicating pregnancy. Methods: Firstly, morning urine of thirty patients with hypertensive disorder complicating pregnancy( including 10 gestational hypertension cases,10 mild pre-eclampsia cases and 10 severe pre-eclampsia) and ten healthy pregnant women were collected. Two-dimensional gel electrophoresis (2-DE) was used to screen the differential proteins,the 2-DE results was analyzed by Image master 2D software,and finally the differentially expressed proteins was identified by means of MALDI-TOF-MS mass spectrometry identification. Results:Comparing to the control group, 65 proteins occurred a significant expression change in the hypertensive disorder complicating pregnancy group,30 differentially expressed proteins were identified. We found 22 gestational hypertension related pro- teins,among which there were 14 up-regulated proteins and 8 down-regulated proteins,26 mild preeclampsia related proteins,among which there were 5 up-regulated proteins and 21 down-regulated proteins,and 25 severe preeclampsia related proteins,among which there were 3 up-regulated proteins and 22 down-regulated proteins. Conclusions: Urinary proteomics is a successful tool to screen differentially expressed proteins of hypertensive disorder complicating pregnancy,which might be related to the occurrence and development of hypertensive disorders in pregnancy. Urinary proteomics providing the basic information for the study of urine specific biomarkers of hypertensive disorder complicating pregnancy.
