摘要
目的:探讨突变的膜结合型 TNF-α基因对人胃癌细胞生长的影响.方法:用突变的 膜结合型 TNF-α基因作为目的基因,以逆转录病毒为载体将目的基因导 入人胃癌细胞 MGC803,经 G418筛选得到抗性克隆 MGCT803结果: Southern杂交证实目的基因已整合在 MGCT803细胞中.用 ELISA方法检测MGCT803细胞表面TNF- α表达量明显增 高.MGCT803细胞形态、体外增殖能力无显著改变.裸鼠体内接种表明,MGCT803细胞在裸鼠体 内的生长受到明显抑制.结论:膜结合型TNF-α基因导入人胃癌细胞后其动物体内致瘤 性明显下降.
Objective: The effects of mutant membrane bound TNF-α gene on the growth of the human gastric cancer line are studied. Methods: The recombinant retroviral vector with mutant membrane bound TNF-α gene were used to transduce the human gastric cancer line, MGCs803 cells. The resistant colonies named as MGCT803 were obtained by G418 screening. Results: The result of southern blot assay showed that the target gene had integrated into the genomic DNA of MGCT803 cells. Cell surface expression of TNF-α as determined by ELISA analysis revealed that the level of TNF-αexpression on MGCT803 cells surface was markedly increased. There was no variance in the morphological appearance and growth rate in vitro of MGCT803, cells. The results of inoculation in nude with these cells indicated that animals bearing MGCT803 cells showed a considerable decrease in size of tumors. Conclusion: the membrane bound TNF-α could suppress tumorigenicity of human gastric cancer cells.
