大鼠脂肪源性干细胞和骨髓间充质干细胞增殖速度的差异性研究

摘要 目的比较大鼠脂肪源性干细胞(ADSCs)和骨髓间充质干细胞(BMSCs)的增殖速度,探讨ADSCs替代BMSCs的可行性。方法从成年雄性SD大鼠取脂肪组织和骨髓分别提取ADSCs和BMSCs,利用3-(4,5-二甲基噻唑-2)-2,5-二苯基四氮唑溴盐(MTT)法检测细胞增殖速度。结果第二代ADSCs和BMSCs细胞接种时光密度(OD)平均值分别为0.028和0.030,两者OD值进行比较差异无统计学意义(t=0.88,P>0.05),于接种后的第1、2、3、4日ADSCs的OD值都显著高于BMSCs(P<0.05)。结论大鼠ADSCs增殖速度显著高于BMSCs。 Objective To compare the difference on the proliferation speed between adipose-derived stem cells( ADSCs )and bone mesenchymal stem cells (BMSCs)in rats, and discuss the feasibility of ADSCs substituting bone mesenchymal stem cells. Methods The ADSCs and BMSCs was extracted from the adipose tissue and bone marrow from adult male SD rats. The cell proliferation speed was detected by MTT method. Results Average of optical density ( OD ) of second generation ADSCs and BMSCs was 0. 028 and 0. 030,respectively, the difference was not statistically significant ( t = 0. 88,P 〉 0. 05 ), OD of ADSCs was significantly higher than that of BMSCs after vaccinating for 1,2,3,4 days with statistically signifieant difference( P 〈 0. 05 ). Conclusion The proliferation speed of ADSCs is significantly higher than that of BMSCs in rats.
