川芎嗪对大鼠脑缺血再灌注后血脑屏障的保护作用及Claudin-5表达的影响 被引量:18

Effects of Ligustrazine on Blood-brain Barrier and Claudin-5 Expression in Rat after Cerebral Ischemia and Reperfusion

摘要 目的研究川芎嗪对大鼠脑缺血再灌注(I/R)后血脑屏障(BBB)通透性及紧密连接相关蛋白Claudin-5表达的影响。方法采用线栓法制作大鼠脑I/R模型,采用伊文斯蓝(EB)法检测BBB的通透性,免疫组化、RT-PCR及Western blot测定缺血再灌注脑组织Claudin-5的表达。结果再灌注3 h和72 h组缺血侧脑组织EB含量均显著高于假手术组(P<0.05),川芎嗪治疗组缺血侧脑组织EB含量均显著低于同时间点再灌注组(P<0.05)。再灌注3 h和72 h缺血侧脑组织Claudin-5表达显著低于假手术组(P<0.05),川芎嗪治疗组缺血侧脑组织Claudin-5表达显著高于同时间点再灌注组对照组(P<0.05)。结论大鼠脑I/R后BBB损伤与Claudin-5表达下调密切相关,川芎嗪可能通过上调Claudin-5的表达恢复紧密连接的功能,降低I/R后BBB的通透性,从而减轻I/R后脑水肿。 Objective To study the in vivo effects of ligustrazine(TMP) on blood-brain barrier (BBB) and expression of Claudin-5 in rats after brain ischemia-reperfusion (I/R). Methods The focal cerebral I/R rat model of middle cerebral artery occlusion ( MCAO ) was estab- lished by intraluminal suture. The change of the permeability of BBB was detected using Evans blue assay. The expression of Claudin-5 was determined by immunohischemistry assay, RT-PCR and Western blot. Results The BBB permeability was found significantly decreased at 3 h and 72 h after I/R compared with sham-operated group. The BBB permeabihty in ligustrazine group was found significantly increased com- pared with the group of I/R at the same time point.s. The expression of Claudin-5 was found significantly decreased at 3 h and 72 h after I/R group compared with sham-operated group. The expression of Clandin-5 in ligustrazine group was found significantly increased compared with the I/R group at the same testing time points. Conclusions The down-regnlation of Claudin-5 is closely related to the BBB injury induced by cerebral ischemia-repeffusion. Ligustrazine contributes to the cerebral edema after reperfusion through the up-regulation of Claudin-5, re- covery of function of tight junction and reducing the permeability of BBB.
