摘要
采用水提醇沉法从绞股蓝中提取、分离纯化获得一种均一多糖(GPP),并对其理化性质及体外抗氧化活性进行了研究。选用大孔树脂和DEAE-52离子交换纤维素对水提醇沉获得的绞股蓝粗多糖进行分离纯化,以HPLC、红外光谱法等对获得的均一多糖的纯度、光谱特性、单糖组成等多种理化性质进行了测定,并通过测定羟自由基、ABTS清除能力以及亚铁离子螯合能力对其体外抗氧化活性进行了初步研究。绞股蓝多糖GPP由甘露糖、葡萄糖醛酸、半乳糖以及少量的木糖和鼠李糖组成,端基碳为α构型,其具有显著的羟自由基、ABTS清除能力以及亚铁离子螯合能力,可以用来作为一种潜在的抗氧化剂。
In order to investigate the physicochemical characteristics and in vitro antioxidant activity of polysaccharide from Gynos-temma pentaphyllum, a homopolysaccharide, named GPP,was extracted and purified from the whole-plant of Gynostemma pentaphyllum through hot water extraction followed sequentially by macroporousresin and DEAE-52 treatment. The physicochemical characteristics of GPP, such as purity, spectral properties and monosaccharide composition, were investigated using methods like HPLC,infrared spectroscopy, ultraviolet spectrum method and so on. The in vitro antioxidant activity of GPP was also evaluated with hydroxyl radical, by ABTS scavenging capacity and iron chelating ability assay. The experiment results showed that GPP was a kind of acidic polysaccharides and no obvious absorption peaks were observed in ultraviolet spectrum at 260 nm and 280 nm. The elemental analysis result suggested that GPP contained no nitrogen, and the proportion of carbon and hydrogen elements is 6.1 to 1. It was showed that a single symmetry peak in the plot of HPLC existed,and the purity was more than 98.8% ,which implied a homogenous polysaccharide. The molecular weight of GPP was 2.52 x 103 k by GPC method. It contained glucuronic acid, mannose, galactose, with little of xylose and rhamnose, measured by PMP derivatization HPLC method. Infrared spectrum analysis showed that carbon end group of GPP was alpha configuration. It was concluded that GPP was composed of mannose, glucuronic acid, and galactose, with a few of xylose and rhamnose. Antioxidant activity in vitro was also studied with the hydroxyl radical, by ABTS scavenging capacity, iron chelating ability assay. The in vitro antioxidant tests showed that GPP had significant inhibition effects on hydroxyl, ABTS radicals and iron chelating capacities in a dose-dependent manner,which provided a theoretical basis for the exploitation of a potential natural antioxidant.
