过氧化氢诱导产蛋鸡原代肝细胞氧化应激模型的建立 被引量:15

摘要 不同浓度过氧化氢(H2O2)作用产蛋鸡肝细胞1、2、4 h和24 h,通过测定细胞相对存活率、胞内丙二醛(MDA)和活性氧(ROS)含量、超氧化物歧化酶(SOD)和过氧化氢酶(CAT)活性,探索H2O2引起产蛋鸡肝细胞氧化应激的合适处理时间及浓度,构建原代肝细胞氧化应激模型。结果表明:4 mmmol/L H2O2、处理肝细胞2 h,与对照组比细胞存活率显著降低(P<0.05),细胞内MDA和ROS产生量显著升高(P<0.05),同时SOD和CAT酶活也升高(P<0.05)。综上所述,4 mmmol/L H2O2、处理肝细胞2 h,可构建产蛋鸡原代肝细胞氧化应激模型。
