摘要
目的研究复元胶囊及其与甲氨蝶呤(MTX)联用对气虚血瘀证类风湿关节炎(RA)的治疗机制。方法用Ⅱ型胶原诱导大鼠RA模型后,采用疲劳、饥饿、高脂饮食及寒湿等方法建立气虚血瘀证模型大鼠,设空白对照组、模型组、复元胶囊组、MTX组、复元胶囊联用MTX组(联用组)。各组以相应的药物灌胃,于8 w后,用酶联免疫吸附试验法(ELISA)检测血清肿瘤坏死因子α(TNF-α),放射免疫法检测血清白细胞介素-1β(IL-1β),白细胞介素-6(IL-6)水平。结果与模型组相比,复元胶囊组、MTX组及联用组大鼠关节肿胀明显减轻,血清TNF-α、IL-1β、IL-6均有所下降(P<0.05),其中以联用组较优(P<0.05)。结论复元胶囊联用MTX可明显改善气虚血瘀证RA大鼠的症状,其作用机制可能与抑制大鼠TNF-α、IL-1β、IL-6等炎症介质的表达有关,由此为复元胶囊治疗气血血瘀证RA大鼠提供了科学依据。
Objective To observe the effect of Fuyuan capsule and Fuyuan capsule united Methotrexate on rhemnatoid arthritis (RA) rats with qi-deficiency and blood stasis syndrome. Methods After established RA models with Collagen Ⅱ, the rat models with qi-deficiency and blood stasis syndrome were induced by fatigue, starvation, high-fat diet and cold. The models were randomly divided into 5 groups: nor- real,model, Fuyuan capsule, methotrexate (MTX) and Fuynan capsule united MTX (combination) groups. The rats were given drugs by gas- tric perfusion for 8 weeks, then the levels of serum tumor necrosis factor alpha (TNF-α), interleukin 1β(IL-1β) and interleukin-6 (IL-6) were measured. Results Compared with the model group, the serum expression of TNF-α,IL-1β, IL-6 were reduced significantly in the Fuyuan capsule, MTX and combination groups (P 〈 0. 05). The combined group was outstanding (P 〈 0. 05). There was no statistically signif- icant difference between Fuyuan capsule and MTX group (P 〉 0. 05 ). Conclusions Fuyuan capsule united MTX can obviously improve the syndromes of RA rats with qi-deficieney and blood stasis syndrome. The mechanism might be related to the drugs inhibiting the expressions of TNF-α,IL-1β, IL-6 and other inflammatory mediators, which provides scientific proofs for the RA treatment with Fuyuan capsule.
