摘要
目的:探讨黑骨藤乙醇提取物对CIA大鼠的治疗作用及作用机制。方法:采用牛Ⅱ型胶原诱导SD大鼠,构建CIA大鼠模型,设置正常组、模型组、阳性组及黑骨藤高、中、低剂量组;观察大鼠踝关节肿胀情况,并进行AI评分,采用HE染色观察踝关节的病理形态变化,并采用酶联免疫吸附法(ELISA)测定大鼠血清中细胞炎症因子IL-6、TNF-α及免疫球蛋白lgA、lgM、lgG的含量变化。结果:与模型组比较,黑骨藤乙醇提取物对CIA大鼠的足关节肿胀表现出良好的抑制作用,能明显降低其AI评分,各治疗组的炎症细胞浸润、滑膜及血管的增生均减轻,显著降低大鼠血清中的IL-6、TNF-α、lgA、lgM及lgG表达水平。结论:黑骨藤乙醇提取物能有效改善CIA大鼠关节炎症状,降低踝关节肿胀度及关节炎指数,其机制可能与血清中炎症细胞因子及免疫球蛋白表达水平下调有关。
Objective:To explore the therapeutic effect and mechanism of the ethanol extract of Periploca forrestii Schltr.on collagen induced arthritis(CIA)rats.Methods:The CIA model was established by immunizing SD rats with bovine typeⅡcollagen.The CIA rats were randomLy divided into 6 groups:normal group,model group,positive group,Periploca forrestii Schltr low-dose group,Periploca forrestii Schltr middle-dose group and Periploca forrestii Schltr high-dose group.After treatment,the swelling of the ankle joints in all animals was assessed and scored.Ankle joints were harvested from rats in each group,and the pathological changes of synovial tissues were observed under light microscope following hematoxylin and eosin(HE)staining.The serum of the rats was collected,and the levels of inflammatory cytokines(IL-6 and TNF-α)and immunoglobulins(lgA,lgM and lgG)were determined by enzyme-linked immunosorbent assay(ELISA).Results:As compared with the model group,the ethanol extract of Periploca forrestii Schltr.significantly suppressed the joints swelling of CIA rats showing lower arthritic scores.Histological pathology demonstrated that the ethanol extract of Periploca forrestii Schltr markedly ameliorated the infiltration of inflammatory cells,hyperplasia of synovium and blood vessels in the ankle joints of CIA rats.ELISA results showed that the levels of IL-6,TNF-α,lgA,lgM and lgG were markedly decreased after treatments of the ethanol extract of Periploca forrestii Schltr.Conclusion:The ethanol extract of Periploca forrestii Schltr.can effectively improve the arthritis symptoms and reduce the joints swelling and arthritis index of CIA rats.The mechanism may be related to the down-regulation of cytokines and immunoglobulins.
