摘要
目的探讨高脂血症病理条件对脑缺血大鼠脑皮质MMP-2和神经元凋亡的影响。方法线栓法建立大鼠大脑中动脉缺血模型(MCAO),免疫组化法检测大鼠脑皮质缺血边缘区MMP-2、Bcl-2和Bax的表达。结果与正常及假手术组相比,MMP-2在单纯脑缺血组、高脂血症组和复合脑缺血组的表达显著增多(P<0.05);与高脂血症及高脂血症假手术相比,复合脑缺血组的MMP-2的表达也是明显增多的(P<0.05),且MMP-2在复合脑缺血7 d组的表达显著高于在单纯脑缺血组的表达。Bcl-2、Bax在伴有脑缺血模型各时间点均有高表达,复合脑缺血组和单纯脑缺血组与相应假手术组组内比较,差异显著(P<0.05);复合脑缺血组与单纯脑缺血组比较,表达下降,缺血后3、7 d组间差异显著(P<0.05);Bcl-2/Bax比值升高,在缺血后7 d组间差异显著(P<0.05)。结论高脂血症的病理基础条件下,MMP-2表达的增加可能对脑缺血后神经元有修复和保护作用。
Objective To study the effect of hyperlipidemia on the expression of MMP-2 and neuronal apoptosis in brain cortex fol- lowing cerebral isehemia of rats. Methods The middle cerebral artery occlusion (MCAO) models were established by intraluminal suture occlusion method, the expressions of MMP-2, Bax and Bcl-2 proteins in the fringe areas of cortex afer cerebral ischemia were detected by im- munohistochemical staining. Results Compared to that of normal control and sham operation groups, the expression of MMP-2 was in- creased in single cerebral ischemia, hyperlipidemia and hyperlipidemia concurrent cerebral ischemia groups, there were significant difference ( P 〈 0. 05 ). Compared to that of hyperlipidemia and sham operation group with hyperlipidemia, the expression of MMP-2 was also dramati- clly increased in 7 days after ischemia with hyperlipidemia group ( P 〈 0.05 ). Moreover, the expression of MMP-2 in ischemia with hyperlip- idemia group was very higher than that of the single cerebral ischemia group. The expressions of Bcl-2 and Bax were high in 3 days and 7 days of cerebral ischemia models, compared to those of sham operation group, there were significant difference between hyperlipidemia con- current cerebral isehemia group and cerebral ischemia group (P 〈 O. 05). In addition, in hyper/ipidemia concurrent cerebral ischemia group and cerebral ischemia group, the expressions of Bax in 3 days and 7 days after ischemia were prominently decreased ( P 〈 0. 05 ) , the ratio of Bcl-2/Bax was increased with significant difference in 7 days after cerebral ischemia ( P 〈 0. 05 ). Conclusions The increased expression of MMP-2 under condition with hyperlipidemia has repaired and protective effect on neurons after cerebral ischemia.
