摘要
DNA甲基化修饰是一种重要的表观遗传修饰,对基因表达调控具有重要影响作用。在哺乳动物中,DNA甲基化主要发生在CpG岛,其通过影响染色质结构、DNA构象、稳定性以及与蛋白质相互作用方式等来发挥作用,调控基因的表达。在转基因动物中,外源基因的表达与DNA的甲基化密切相关,CpG岛甲基化介导的基因沉默往往使外源基因不表达或低表达。而在人类基因组中异常的DNA甲基化常常伴随着疾病的发生。本文通过对哺乳动物中CpG岛甲基化研究现状及甲基化调控基因表达可能的分子机制进行了综述,并对其发展趋势及应用前景进行了展望。
DNA methylation is an important epigenetic modification, which has an important effect on the regulation of gene expression. It plays the role on gene expression by changing chromatin structure, DNA conformation and stability as well as protein -protein interaction mode. Exogenous gene expression and DNA methylationare closely related in the transgenic animals and CpG island methylation - mediated gene silencing often lends to no or low expression of the exogenous gene. In the human genome, the abnormal DNA methylation is often accompanied by the occurrence of the disease. This review focuses on the molecular mechanism between DNA methylation and gene regulation in mammal.
