摘要
以苹果渣粗提物为原料,用大孔吸附树脂分离根皮苷,以高效液相色谱法测定根皮苷含量.利用Box-Behnken中心组合设计原理,应用响应面试验优化方法确定大孔吸附树脂最优分离条件.试验结果显示,最优吸附条件为:吸附液浓度0.5mg/mL、吸附液pH值4、吸附流速1.0mL/min,在此条件下树脂吸附率可达到69.87%,各因素对吸附率的影响顺序为吸附流速>吸附液pH>吸附液浓度;最优洗脱条件为:乙醇洗脱液浓度80%、洗脱液pH值7、洗脱温度70℃,在此条件下树脂洗脱率可达89.92%,各因素对洗脱率的影响顺序为洗脱液pH>乙醇洗脱液浓度>洗脱温度.
The crude extract from apple pomace was used as raw material, and the phlorizin was extracted from it and was tested by HPLC. The optimization condition of extraction con- dition of macroporous adsorption resin was confirmed using response surface methodology with Box-Behnken design. The result showed that the optimized absorption condition was: the content of absorption solution, pH and flow rate were 0.5 mg/mL, 4, 1.0 mL/min, re- spectively, and in this condition the absorption rate could reach to 69.87%, and the sequence of each factor effecting on absorption rate was: flow rate〉 pH〉the content of absorption liquid; the optimized desorption condition was: the content of desorption ethanol solution, pH and temperature were 80%,and 70 ℃, and in this condition the desorption rate could reach to 89.92%, and the sequence of each factor effecting on desorption rate was: pHi〉 the content of desorption ethanol solution〉 temperature.
