摘要
目的:通过用组织病理学方法结合局部脑血流量(rCBF)测定,对大鼠局灶性脑缺血动物模型缺血半暗带的解剖定位进行初步探讨。方法:用线栓法制成大鼠大脑中动脉(MCA)闭塞及再通模型、采用TTC染色法观察分组动物在MCA闭塞的不同时间再灌流48h后脑梗死灶的分布,并用氢清除法测定缺血区rCBF的变化。结果:大鼠MCA闭塞1~2h,梗死灶主要位于缺血侧的外侧尾壳核和额顶叶皮质下部;MCA闭塞3h、梗死灶向周围扩大;MCA闭塞4h、梗死灶进一步扩展至大部分新在质区,但与MCA闭塞6h时无明显区别。MCA闭塞后缺血侧的内侧尾壳核和额顶叶皮质上部rCBFG下降至对照组的27%~45%。结论:大鼠MCA闭塞后在2~3h的时间窗以内恢复血流,可使位于缺血边缘区的内侧尾壳核和额顶叶皮质上部脑组织被挽救,该区域可能相当于缺血半暗带的等值区。
Aim:To assess the rat localization of penumbral area in a model of focal cerebral ischemia in rat.Methods:The middle cerebral artery(MCA)of Wistar rats was occluded for 1,2.3,4,and 6 h with the use of an intralaminal filament and re-circulation was instituted for 48 h.Regional cerebral blood flow(rCBF)was measured by using the hydrogen clearance method,and using 2,3,5,-triphenylterazolium chloride(TTC)staining to identified infarcted areas.Results:Following 1-2 h of MCA occlusion,infarcted areas localized to the lateral caudoputamen and lower frontoparietal cortex.Prolongation of MCA occlusion to 4 h,led to extension of the infarcts to the caudoputamen and large parts of neocortex,when the infarct size was similar to after 6 h of MCA occlusion,rCBF in the perifocal ipsilateral areas about 27%~45%of control.Conclusion:The re-sults show that the medial caudoputamen and upper frontoparietal cortex can be salvaged by reper fusion so that both the rCBF and the recovery studies with a histopathological endpoint reversible area may be equivalent to the ischemic penumbra in this model.
