不同浓度STZ腹腔注射与2型糖尿病大鼠模型建立 被引量:8

摘要 目的:探讨2型糖尿病(T2DM)大鼠模型的链脲菌素(Streptozocin,STZ)最佳注射浓度。方法:80只8周龄SFP雄性SD大鼠高糖高脂饮食两周后随机分组,分别按40、45、50 mg/Kg体重剂量腹腔注射链脲菌素建立T2DM大鼠模型,同时继续高糖高脂食物;对照组腹腔注射柠檬酸50 mg/Kg,普通饲料喂养。各组为20只。分别在第2、5、10、20、30、45、60天测大鼠的血糖值,以10天后血糖>16.7 mmol/L计算糖尿病成模率,并计算60天内的大鼠死亡率。结果:实验组各组成模率分别是60%、90%、70%,对照组为0,此外,50 mg/Kg剂量组大鼠死亡5只,其他组均为0。结论:45 mg/kg STZ腹腔注射大鼠成膜率高,安全性强,维持时间长,是理想的2型糖尿病造模浓度。 Objective:To find the best dosage of streptozocin (STZ) that was used to induce the rat model of Type 2 diabetes mellitus (T2DM). Methods:80 SPF grade of 8 weeks male SD rats were fed with high glucose and high fat diet for 2 weeks and then randomly divided into 4 groups, STZ (30 mg,/kg, 40 mg/kg, 50 mg/kg) was injected intraperitoneally to make rat model of T2DM. The experimental groups were fed with high glucose and high fat diet continually. The control group was injected citric acid by 50rag/ kg and fed with normal diet. The blood glucose was measured at 2,5,10,20,30,45,60 day, respectively. To calculate the success model rate (blood glucose 〉 16.7 mmol/L after 10 days ) and the mortality rate (within 60 days). Results: The succeed model rate was 60%, 90% , 70% in experimental groups respectively and 0 in control group. 7 rats died in the 50mg/kg group, other groups were 0. Conclusions: The 45mg/kg group has the highest success rate of making model, strongly security and long duration, which is the best dosage of STZ to induce rat model of T2DM.
