摘要
采用荧光原位杂交(FISH)技术研究45S rDNA在10个中国古老月季品种染色体上的差异。结果表明:(1)中国古老月季品种的45S rDNA杂交位点数与染色体倍性变化基本一致,中国古老月季的起源可能比较简单:二倍体的‘月月红’、‘月月粉’和‘绿萼’均有2个45S rDNA杂交位点,三倍体的‘湖中月’、‘青莲学士’和‘春水绿波’有3个45S rDNA杂交位点,四倍体品种除‘大富贵’有3个杂交位点外其他都有4个45S rDNA杂交位点;(2)中国古老月季品种之间在45S rDNA杂交信号的强弱上存在差异,说明中国古老月季品种之间在染色体结构及45S rDNA拷贝数上存在多样性。
The diversity of 45S rDNA ribosomal RNA genes (rDNA) was studied by fluorescence in situ hybridization (FISH) in the chromosomes of 10 Chinese old garden rose cultivars. The results were as following: ( 1 ) Rosa chirtensis ' Slater's crimson China' , R. chinertsis ' Parson's pink China' and R. ' Viridiflora' had two 45S rDNA sites on their diploid genomes. R. ' Huzhongyue' , R. ' Qinglianxueshi' and R. ' Chunshuiltibo' had three 45S rDNA loci on their triploid genomes. Except that only three 45S rDNA sites were observed on the nucliei of R. ' Dafugui' , there were all four 45S rDNA loci on the genomes of tetraploid cultivars, such as R. ' Yipinzhuyi' , R. ' Mudanyueji' and R. ' Jinfenlian'. So, the number of 45 S rDNA sites was almost consistent with the ploidy level of Chinese Old Garden Roses, indicating that the Chinese Old Garden Roses might have a simple origination withonly several wild rose species involved. (2) The intensities of 45S rDNA hybridization signals differed much among the 10 cultivars, suggesting that the Chinese old garden roses were diversified in chromosomal structure and 45S rDNA copies.
