摘要
探讨人参总皂甙(totalsaponinsofpanaxginseng ,TSPG)对人肝癌细胞株HepG2碱性磷酸酶活性及甲胎蛋白生成的影响。方法 :用酶促反应试剂盒检测细胞浆中碱性磷酸酶(alkalinephosphatase,ALP)活性 ,用免疫组化染色和ELISA法定性和定量检测细胞浆中甲胎蛋白(alpha fetoprotein,AFP)合成情况 ,观察TSPG对ALP和AFP的影响。结果 :400μg/mlTSPG使反映肝细胞分化的ALP活性明显升高(P<0 01) ,并使肝癌标志蛋白AFP合成明显下降(P<0 01)。结论 :TSPG具有使HepG2细胞的胞浆中某些表型逆转 。
Objective: To investigate the effects of total saponins of panax ginseng (TSPG) on the enzyme activity and protein synthesis in human hepatoma cell line HepG2 Methods: The specific activity of alkaline phosphatase (ALP) in cytoplasma was assayed by using ALP Kit based on Bessey method Alpha fetoprotein (AFP) in cytoplasma was determined qualitatively and quantitativelyby immunohistochemistry and ELISA Results: After treatment with 400μg/ml TSPG, the specific activity of ALP which reflected liver cell differentiation was significantly increased (P<0 01), while the production of AFP, the liver cancermark protein, was significantly decreased (P<0 01) Conclusions: These results indicated that TSPG could induce reverse changes of some plasmo phenotypes in the HepG2 cells, which could be considered as effects of differentiation induction
