摘要
目的检测LMO4在APP/PS1-Tg转基因小鼠脑组织中的表达。方法选取6月龄大小的APP/SP1-Tg转基因小鼠为实验组,同月龄同种系的野生型小鼠C57BL/6为对照组,分别提取其脑组织海马mRNA和蛋白,应用实时荧光定量PCR和蛋白免疫印迹方法检测两组小鼠脑组织中LMO4蛋白和基因的表达。结果 LMO4蛋白和基因在APP/SP1-Tg转基因小鼠海马组织表达水平明显低于月同种系的野生型C57BL/6小鼠海马组织表达水平(P<0.05)。结论 LMO4在APP/SP1-Tg转基因小鼠低表达可能与阿尔茨海默氏病的发病过程相关。
Objective To detect the expression of LIM-domain only 4(LMO4) in the hippocampus of the APP/PS1 transgenic mouse model of Alzhaimer's disease. Methods Six-month-old APP/PS1-Tg transgenic mice and age-matched wild-type C57BIJ6 mice were used for determing the expression of LMO4 protein and gene in the hippocampus using Western blot and Real-time qPCR. Results The expression levels of LMO4 mRNA and protein in hippocampus were significantly decreased in the APP/PS1-Tg transgenic mice than in wild-type C57BL/6 mice(p〈0.05). Conclusion The decrease of LMO4 expression level in APP/PS1-Tg transgenic mice might be related to the Alzheimer disease development.
