摘要
目的对壳寡糖混合物进行分离纯化,得到结构确定的不同聚合度的单体并对其进行理化性质分析。方法采用聚丙烯酰胺凝胶Bio Gel P-6及P-6Fine色谱柱分离壳寡糖混合物,得到5个组分,运用薄层色谱法(TLC)、高效液相色谱法(HPLC)、质谱法(MS)、红外光谱法(IR)对其进行分析。结果分离得到的5个单体分别为壳三糖、壳四糖、壳五糖、壳六糖和壳七糖。结论利用凝胶过滤色谱法可以分离制备结构确定、不同聚合度的壳寡糖单体。
Objective To isolate and obtain chitooligosaccharide isomers with different degrees of polymer- ization and analyze their physicochemical properties. Methods Five components were isolated and puri- fied by polyacrylamide gel Bio Gel P-6 and P-6 Fine column. The components of each fraction were ana- lyzed by thin layer chromat- ography (TLC), high performance liquid chromatography (HPLC), mass spectrometry(MS) and infrared spectroscopy (IR). Results The five fractions were glucosamine trimer, glucosamine tetramer, glucosamine pentamer, glucosamine hexamer, and glucosamine heptamer respec- tively. Conclusion Polyacryla mide gel Bio Gel P-6 can be used for separating well defined chitooligosac- charide isomers with different degrees of polymerization.
