摘要
目的:探讨青蒿琥酯(Art)影响基质金属蛋白酶-2(MMP-2)的抗肝纤维化作用机制。方法:采用二甲基亚硝胺(DMN)ip 6周,制备大鼠肝纤维化模型。动物随机分为6组:正常对照组、模型对照组、Art(5,15,45 mg.kg-1)组、阳性对照秋水仙碱(Col,0.1 mg.kg-1)。检测肝组织中羟脯氨酸(Hyp)的含量、动物血清透明质酸(HA),层连蛋白(LN),Ⅲ型前胶原肽(PⅢP)水平及肝组织MMP-2 mRNA和蛋白的表达。结果:与模型组相比,Art各组能降低肝组织Hyp含量(P<0.01),同时血清HA,LN,PⅢP,肝组织MMP-2-mRNA和蛋白的表达水平下降(P<0.05,P<0.01)。结论:Art能有效抑制DMN诱导的大鼠肝纤维化及肝脏中MMP-2的表达。
Objective: To investigate the effects of artesunate(Art) on Matrix Metalloproteinase-2(MMP-2) activity, explore the mechanism against liver fibrosis. Method: Liver fibrosis model of rats was induced by injecting N-nitroso dimethylamine (DMN) for 6 weeks. Rats were randomly divided into 6 groups: the normal group, the model group, the positive control group treated by colchicine(0.1 mg·kg^-1·d-1), and the three Art groups(5,15,45 mg·kg^-1·d-1). The content of hydroxyproline (Hyp) of liver tissue, hyaluronic acid (HA),laminin (LN),collagen type Ⅲ peptide (PⅢP) of serum were analyzed. Expressions of MMP-2 protein and mRNA were assayed by Western blot and RT-PCR. Result: The content of Hyp in liver and HA,LN,PIIIP of serum in artesunate group were remarkably decreased and expressions of MMP-2 protein and mRNA were down-regulated compared with the model group. Conclusion: Artesunate can effectively inhibit the progress of hepatic fibrosis and the expression of MMP-2 in rats of liver fibrosis induced by DMN.
