七氟醚预处理对大鼠肺缺血再灌注时肺组织血管紧张素转化酶mRNA表达的影响 被引量:8

Effects of sevoflurane preconditioning on expression of angiotensin converting enzyme mRNA in lung tissues during lung ischemia-reperfusion in rats

摘要 目的探讨七氟醚预处理对大鼠肺缺血再灌注时肺组织血管紧张素转化酶(ACE)mR—NA表达的影响。方法成年雄性sD大鼠54只,体重250~320g,采用随机数字表法,将其分为3组(n=18):假手术组(S组)仅游离左肺门,但不阻断;肺缺血再灌注组(I/R组)采用阻断左肺门45min后再灌注120min的方法制备大鼠肺缺血再灌注模型;七氟醚预处理组(SP组)吸入2.1%七氟醚30min,停止吸入后10min时后制备肺缺血再灌注模型。于再灌注30、60和120min时随机取6只大鼠,处死取肺组织,测定湿,干重比(W/D比),采用比色法测定髓过氧化物酶(MP0)活性,采用RT-PCR法测定ACEmRNA的表达,光镜下观察肺组织病理学结果。结果与S组比较,I/R组和SP组再灌注各时点肺组织W/D比、MPO活性和ACEmRNA表达水平均升高(P〈0.05);与I/R组比较,SP组再灌注各时点肺组织W/D比、MPO活性和ACEmRNA表达水平降低(P〈0.05)。SP组肺组织病理学损伤较I/R组减轻。结论七氟醚预处理可能通过下调ACEmRNA的表达从而减轻大鼠肺缺血再灌注损伤。 Objective To investigate the effects of sevoflurane preconditioning on the expression of pulmo- nary angiotensin converting enzyme (ACE) mRNA in lung tissues during lung ischemia-reperfusion (I/R) in rats. Methods Fifty-four male Sprague-Dawley rats, weighing 250-320 g, were randomly divided into 3 groups ( n = 18 each) : sham operation group (group S), I/R group and sevoflurane preconditioning group (group SP). The an- imals were anesthetized, tracheostomized and mechanically ventilated. Lung I/R was induced by clamping the left pulmonary hilum for 45 min followed by 120 min reperfusion in groups I/R and SP. While the left pulmonary hilum was only isolated but not ligated in group S. 2.1% sevoflurane was inhaled for 30 rain, and lung I/R was induced 10 min after the end of inhalation in group SP. Six rats in each group were chosen at 30, 60 and 120 min of reper- fusion and sacrificed, lungs were removed for determination of wet/dry lung weight (W/D) ratio, MPO activity (by colorimetric method) and the expression of ACE mRNA (by RT-PCR) and for microscopic examination in lung tis- sues. Results Compared with group S, the W/D ratio, MPO activity and expression of ACE mRNA were signifi- cantly increased in groups I/R and SP (P 〈 0.05). Compared with group I/R, the W/D ratio, MPO activity and expression of ACE mRNA were significantly decreased in group SP (P 〈 0.05 ). Microscopic examination showed that the pathologic changes were significantly attenuated in group SP compared with group I/R. Conclusion Sevoflurane preconditioning can reduce lung I/R injury through downing-regulating the expression of ACE mRNA in rats.
