摘要
目的研究人外周血淋巴细胞(PBMCs)体外诱导前后的蛋白质组表达变化,进一步探讨其抗肿瘤机制。方法收集培养前后的细胞提取总蛋白,定量、双向电泳和银染后,利用ImageMasterTM软件对表达异同的蛋白质点进行分析及质谱鉴定。结果培养前后细胞相似的蛋白主要与基因转录因子和细胞骨架相关,差异蛋白主要与细胞凋亡和代谢相关。结论 CIK细胞的扩增及表型变化与细胞内蛋白表达改变相关。
This study aimed to evaluate proteomics differences of human peripheral blood lymphocytes (PBMCs) before and after induced culture in vitro and further explore its antitumor mechanism. Firstly, cell samples beforeand after cultivation were collected, and the total proteins in the samples were extracted, quantitated, and subjected to two-dimensional eleetrophoresis and silver staining. Then the same and different expressions of protein spots wereanalyzed by ImageMasterTM 2D Platinum and identified by mass spectrum (MS). The results showed that similar expressional protein spots mainly related with gene transcriptional factors and cytoskeletons. While different spotsmostly related with cell apoptosis and metabolism. All result suggested that CIK cells' proliferation and change of phenotypes are associated with discrepant protein expression patterns of the cells before and after culture.
