摘要
目的:探讨转录调控因子Twist1对人乳腺癌MCF-7细胞E-cadherin表达的影响。方法:上调人乳腺癌MCF-7细胞中Twist1的表达,利用RT-PCR,Western blot,细胞免疫荧光检测分析上调Twist1对E-cadherin表达的影响;划痕实验分析对细胞侵袭运动能力的影响;三维培养观察Twist1对MCF-7细胞成血管能力的影响。结果:MCF-7细胞过表达Twist1抑制细胞E-cadherin的表达,并且导致其亚细胞定位异常,细胞迁移运动能力增强,细胞成血管能力增强。结论:Twist1能抑制MCF-7细胞E-cadherin的表达,并增强其侵袭迁移及血管生成能力。
Objective:This study aims to investigate the effect of Twist1 on human breast carcinoma E-cadherin. Methods: pcD-NA-Twist1 was transfected into human breast carcinoma cell line MCF-7. The expression of E-cadherin was detected via real-time polymerase chain reaction and immunofluorescence staining. Wound-healing assay was performed to examine the migration ability of cells, and the 3D culture was used to observe vasculogenic mimicry (VM) formation. Results:MCF-7 cells overexpressing Twist1 resulted in the suppression of E-cadherin expression and abnormal subcellular location. Migration ability and VM formation were also enhanced. Conclusion:Twist1 prevented E-cadherin expression as well as promoted migration ability and VM formation in human breast carcinoma cell line.
