摘要
用移植人小细胞肺癌的裸鼠作为肿瘤模型,研究了碘标记单克隆抗体2F7在体内的肿瘤显fi,抗体、药物偶合物的连接,导向药物2F7-HSA-MTX的体外细胞毒及体内肿瘤抑制作用。结果显示,碘标记抗体在荷瘤小鼠体内有确切的肿瘤定位作用,清晰的肿瘤显像持续到168h以后。间接连接的导向药物可携带更多的MTX分子,细胞毒作用比直接连接提高2倍。导向药物可使荷瘤小鼠的长瘤潜伏期延长,长瘤率降低,抑瘤率为84%和自然生存期延长。
Using nude mice bearing human small cell lung cancer (SCLC) as a model of tumor, radioimmunoimaging with monoclonal antibody 2F7 against SCLC and immu-notargeting chemotherapy in vitro and in vivo were studied, it was demonstrated in the imaging test that 125 I-labeled 2F7 retained in tumor more than 168 H and the clear cut Y camera pictures could be obtained In nude mice bearing the zenografts. Compared with 2F7-MTX the drug-carrier-antibody conjugate could carry MTX molecule more than 20 fold and cytotoxicity more than two fold.The results of exper mental therapies showed that the appearance of tumor was delayed, the tumor incidence rate was lower, the inhibition rate of tumor growth (%IC) was 84% and the life span was longer in the targeting therapy groups, compared with the controls
