抗人CD_3、CD_7—PAPs免疫毒素对T细胞的联合杀伤效应

摘要 免疫毒素(IT)杀伤试验分8组进行。主要采用改进的MTT法测定不同浓度的抗CD3-PAPs、抗CD7-PAPs、抗CD3-PAPS十抗CD7—PAPs、多抗AHTG—PAPs杀伤T细胞的效率,同时以B细胞作非靶细胞对照。结果证实,两种抗人T细胞单抗IT和多抗IT均具有特异性杀伤T细胞的作用。比较杀伤率发现,单抗IT特异性高,杀伤力也较强;多抗IT杀伤力强,但特异性较差;而两种单抗IT合用则杀伤力进一步提高,并能保持其特异性。结果提示,我们制备的两种单抗IT可用于体夕P清除T细胞作异体骨髓和/或脐带血移植;两者联合应用可能其协同作用效果更好。 There were eight groups in the cytotoxicity test, i. e. anti-CD3-PAPs,anti--CD7-PAPs, anti-CD3-PAPs + anti-CD7-PAPs, AHTG-PAPs, NIgG--PAPs,antiCD3 McAb,anti-CD3 McAb+PAPs and free PAPs. Each was separately added into T and B lymphocytes,cultured in 96-well plates. An improved MTT assay was performed,as references to determine the efficacy and specificity of immunotoxins. The results showed that the groups of anti--CD3-PAPs,anti--CD7--PAPs and AHTG-PAPs had a selective cytotoxicity to T lymphocytes. McAb immunotoxins showed a higher specificity but lower cytotoxicity than PcAb immunotoxin. The mixture of antiCD3-PAPs with anti-CD7-PAPs showed a similar cytotoxic efficacy and higher specificity compared with AHTG-PAPs. The results indicated that the combination o f McAb immunotoxins would be more effective than any one alone in the targeting therapy,especially in bone marrow or umbilical cord blood transplantation as purging reagents.
