摘要
为了有效地控制细胞质量、我们从1987年开始、相继建立了抗口腔支原体(M.orale),精氨酸支原体(M.arginini),猪鼻支原体(M.hyorhinis).和莱氏无胆甾原体(A.laidlawii)的杂交瘤细胞系共20株。并用IFA法、BA法或APAAP法对单克隆抗体进行了检测和鉴定。单克隆抗体(McAb)亚类鉴定结果:8株为IgG1,2株为IgG2b,其余10株为IgM。抗体效价测定大多数在4000~8 000之间。利用支原体纯培养的菌落进行了IFA染色试验,封闭试验,生长抑制试验和免疫电镜检查,均证实了它们的特异性,并获得2株对同属支原体有抗原交叉的McAb(3D1和3A11)。利用上述McAb检测实验室常用细胞的支原体污染,较常规法和DNA荧光染色法简便,快速,特异性较高。
Twenty hybridoma cell line secreting monoclonal antibodies against mycoplas-ma was obtained. They were specific McAb against M. orale, M. arginini , M. hyorh-inis and A. laidlawii respectively. A mean of the chromosome of these hybridoma cells was 104 classesor subclasses of these McAbs were IgG1 for 8 lines, IgG2b for 2 lines, IgM for 10 lines. The titer of these McAb was 1 :400-8 000, using immunoassay of IFA, BA and APAAP. The specificity of these antibodies was demonstrated by IFA, block test, growth inhibition test and immunoelectromicroscopy.3D1 and 3A11 hybridoma cell lines were specific. We applied these McAbs to detect mycoplasma in cell culture and beast speciments and obtained satisfictory results.
