摘要
以大苞白山茶叶片为外植体,研究不同生长调节剂6-BA、TDZ、KT、2,4-D和NAA对其愈伤组织诱导的影响。结果表明:MS+3.0 mg/L 6-BA+0.5 mg/L NAA愈伤诱导率最高,但以MS+2.5 mg/L 6-BA+0.5 mg/L NAA愈伤组织分化最好;在6-BA 2.5 mg/L+NAA 0.5 mg/L培养基中分别加入2,4-D(0.5~2.5 mg/L)、KT(0.5~2.5 mg/L)和TDZ(0.1~0.5 mg/L)后,2,4-D和TDZ均可提高愈伤组织的诱导率,最高可达100%和94.8%,KT的加入反而降低了愈伤组织的形成;适宜的分化培养基为MS+2.5 mg/L 6-BA+0.02 mg/LTDZ+0.05 mg/L NAA。
The young leaf of Camellia granthamina were used as explants in the tissue culture research. The effects of different combination of 6-BA, TDZ, KT, 2,4-D and NAA on callus induction were investigated. The results indicated that the appropriate medium for callus induction was MS+2.5 mg/L 6-BA+0.5 mg/L NAA ;MS+6-BA 2.5 mg/L + NAA 0.5 mg/L medium respectively supplied with 2,4-D 0.5-2.5 mg/L, KT 0.5-2.5 mg/L, TDZ 0.1-0.5 mg/L, 2,4-D and TDZ improved callus induction rate up to 100% and 94.8%, KT reduced callus induction; the appropriate medium for differentiation was MS+ 2.5 rng/L 6-BA + 0.02 mg/L TDZ + 0.05 mg/L NAA.
